Composition
Each 5 ml ampoule of Magnerot 500 contains 500 mg of Magnesium Orotate. Magnesium orotate is a bioavailable form of magnesium combined with orotic acid, which supports cellular metabolism and cardiovascular health. Inactive ingredients include water for injection and stabilizing agents.
Description
Magnerot 500 is a parenteral magnesium preparation used for the rapid correction of magnesium deficiency. Administered via intramuscular or slow intravenous injection, it bypasses gastrointestinal absorption and ensures immediate therapeutic availability in cases where oral magnesium is ineffective or contraindicated.
Indications
- Treatment of clinically diagnosed magnesium deficiency
- Supportive therapy in cardiovascular disorders such as arrhythmia and hypertension linked to magnesium imbalance
- Correction of deficiency due to alcoholism, malnutrition, or gastrointestinal losses (e.g., chronic diarrhea or malabsorption)
- Adjunct therapy in pregnancy-related complications including eclampsia and preterm labor prevention
- Prevention of magnesium deficiency during prolonged parenteral nutrition or diuretic use
Contraindications
- Severe renal impairment with risk of hypermagnesemia
- Myasthenia gravis
- Adrenal insufficiency (e.g., Addison’s disease)
- Hypersensitivity to magnesium orotate or any excipients in the formulation
Dosage and Administration
- Route of administration: Intramuscular (IM) or slow intravenous (IV) injection
- Adults: Typically 1–2 ampoules per day (500–1000 mg magnesium orotate), depending on severity
- Children: Dosing based on weight and clinical condition (under supervision)
- Rate: IV administration must be performed slowly to avoid hypotension or cardiac effects
- Monitoring: Regular monitoring of serum magnesium levels is recommended during prolonged use
Precautions and Warnings
- Use cautiously in patients with mild to moderate renal insufficiency
- Avoid rapid IV administration to prevent hypotension, bradycardia, or respiratory depression

Adverse Effects
- Common: Flushing, warmth, sweating, mild hypotension

Drug Interactions

Overdose
Symptoms of magnesium overdose may include:

- Flaccid paralysis
- Respiratory depression
- Cardiac arrest (in extreme cases)
Treatment involves cessation of magnesium administration and intravenous administration of calcium gluconate as an antidote. Supportive measures may include mechanical ventilation and renal dialysis.
